She was born on April 4, 1960, and she began her acting career in theatre before making significant performances in films such as Breaking In (1989), Hudson Hawk (1991), and Dangerous Minds (1995). As a lead actress, some of her acclaimed roles include Rene Jackson in the Lifetime television drama series Any Day Now and defense attorney Shambala Green in the NBC legal drama Law & Order. She later joined the casts for Crossing Jordan and Saving Grace. Toussaint's acting caliber brought her the Black Reel Award, a Critics' Choice Television Award, and a Screen Actors Guild Award.

She received praise and an Independent Spirit Award nomination for her role in the 2012 drama film Middle of Nowhere, by Ava DuVernay. In 2014, she portrayed Yvonne "Vee" Parker, the main antagonist in the second season of the Netflix comedy-drama series Orange Is the New Black. The role earned her a Critics' Choice Television Award for Best Supporting Actress in a Drama Series. She also featured in Selma, a 2014 historical drama film. Later in her career, she co-starred in Forever, Rosewood, Into the Badlands, The Village, and The Equalizer. She also appeared in the films Fast Color, Scary Stories to Tell in the Dark, The Glorias, and Concrete Cowboy.
