In the heart of rural Kansas, a lovable Great Dane named Marmaduke (teenage years, none the wiser) shares his home with Carlos, a sassy feline roommate. His owner, Phil Winslow (the voice of reason), toils away for Bark Organic dog food, leaving little room for Marmaduke's mischief. To Marmaduke, Phil is as stern as a librarian on a mission to silence the library.

The tranquility of their Kansas life is disrupted when Carlos whispers secrets in Marmaduke's ear: they're being transferred to Orange County, California! As they settle into their new home, Phil receives an assignment from his boss, Don Twombly (the mastermind behind Bark Organic's expansion), to create an ad campaign that will make Petco swoon. Phil and Don converge at the dog park, where Marmaduke meets Mazie, a charming canine who develops a crush on our hero. He also encounters Giuseppe, a Chinese Crested dog consumed by fear, and Raisin, a pint-sized Dachshund with an IQ to match her diminutive stature.

As Marmaduke navigates this new world, he crosses paths with Jezebel, a stunning Rough Collie, whose boyfriend, Bosco, a menacing Beauceron, rules the roost with his Miniature Pinscher minions, Thunder and Lightning. Bosco's intimidation tactics leave Marmaduke quaking in his paws.

Under cover of darkness, Marmaduke and the misfit mutts crash a high-stakes party hosted by Bosco, only to be chased away by the would-be king of Orange County. Marmaduke turns to Mazie for advice on winning over Jezebel, but she assumes he's talking about her – little does Marmaduke know that his true target is the beautiful Rough Collie.

In a desperate bid to boost his popularity, Marmaduke concocts a plan with Carlos: stage a fake fight in the dog park, complete with dramatic flair and a hint of showmanship. The stunt works wonders for Marmaduke's image, but his newfound fame comes at a cost – he enters the dog surfing contest, organized by Phil as a promotional ploy to woo Petco, and emerges victorious, much to the dismay of Bosco, the reigning champion.

As tensions between Marmaduke and Bosco escalate into a full-blown brawl, the Petco executives are appalled. In response, Phil hires Anton, a dog trainer with a questionable track record, to corral Marmaduke's energy – with limited success, at first.

As Marmaduke's world begins to crumble, he orchestrates a disastrous dog park soiree that leaves his friends fleeing for cover. Meanwhile, Jezebel (proudly) observes from a distance as Marmaduke (played by) unwittingly sabotages his own social standing. The Winslow clan, oblivious to the chaos unfolding behind them, returns to their boat, only to discover Phil's cunning plan to use their time away to perfect the Petco pitch.

Phil's underhanded tactics are soon exposed when Bosco crashes the party and uncovers Marmaduke's true identity. As the truth comes to light, Marmaduke's friends abandon him, leaving him with a ravaged home and a shattered sense of self. A forlorn Marmaduke slinks away, abandoning his prized possessions, including a cherished toy gifted by Mazie (played by). Unbeknownst to Marmaduke, Mazie embarks on a desperate search for her wayward companion.

Marmaduke's journey takes an unexpected turn as he encounters Chupadogra, a wise and enigmatic English Mastiff with a reputation for devouring his owner. In reality, this grizzled veteran had fled to lead the pack but was subsequently abandoned. As Marmaduke seeks guidance from the elderly canine, Buster implores him to return home to his family while he still has one. With Buster's blessing, Marmaduke sets out on a solo adventure, only to find himself lost in the wilderness.

The Winslows, meanwhile, begin to frantically search for their missing pet, only to stumble upon Mazie (played by) and Marmaduke (played by) simultaneously. However, fate has other plans as the street beneath them collapses, sending Mazie plummeting into a subterranean rainwater conduit. Unhesitatingly, Marmaduke follows suit, prompting Phil to launch a daring rescue mission.

As events unfold, Phil's misstep with Petco proves costly, leading to his termination. Undeterred, he launches an all-out effort to save Marmaduke, who finds himself swept away by the raging waters. In a heart-stopping climax, Phil begs Marmaduke to release his grip on a flimsy branch and surrender to the torrent. Reluctantly, Marmaduke complies, allowing Phil to reel him in.

The internet sensation that ensues – a YouTube video garnering 682,000 views – ultimately secures Phil's rehiring and Bark Organics' partnership with Petco. In the aftermath, Phil begins to contemplate a return to his Kansas roots, but the Winslow family remains resolute in their decision to remain in California, surrounded by the sun-kissed beauty they've grown to love.

As Marmaduke (voiced by Owen Wilson) asserts his egalitarian views, he boldly confronts the self-proclaimed elite canine clique, emphasizing that breed disparities should not dictate their access to the park. The once-fragmented group swiftly unites behind his vision of unity, abandoning Bosco's divisive influence after he confesses his irrational fear of bees - a phobia shared by Marmaduke himself.
