The cinematic chaos that ensues in Scary Movie 2 commences with a sidesplitting spoof of The Exorcist, as Megan Voorhees (Natasha Lyonne) becomes the unwitting vessel for a malevolent force. Two bemused priests, Father McFeely (James Woods) and Father Harris (Andy Richter), attempt to perform an exorcism, but their efforts are met with a series of comically gruesome vomiting fits instead. The ritual goes awry when Megan's sarcastic remarks about McFeely's mother prompt him to take drastic action, resulting in her untimely demise.

One year has passed since the events of the first film, and Cindy Campbell (Anna Faris), Brenda Meeks (Regina Hall), Ray Wilkins (Shawn Wayans), and Shorty Meeks (Marlon Wayans) are attempting to leave their dark pasts behind as they navigate college life. Although all four characters met their demise in Scary Movie, a deleted scene reveals that Brenda's death was merely a near-death experience, while Cindy's status remains unclear. As they struggle to move forward, the group is unwittingly thrust into a series of bizarre and hilarious misadventures.

The story takes a turn when Professor Oldman (Tim Curry), a charismatic but eccentric educator, and his trusty sidekick Dwight (David Cross), a wheelchair-bound research assistant, concoct a plan to study paranormal activity at the notorious Hell House. They recruit a group of unsuspecting college students, including Cindy, Brenda, Ray, Shorty, Alex Monday (Tori Spelling), Tommy (James DeBello), and Buddy (Christopher Masterson), as unwitting test subjects. Theo (Kathleen Robertson), a newcomer to the group, soon finds herself entangled in their antics.

As the group settles into the eerie confines of Hell House, they encounter a cast of eccentric characters, including a foul-mouthed Amazon parrot and Hanson (Chris Elliott), the mansion's peculiar caretaker, who sports a genetically abnormal hand. The night is filled with an assortment of wacky mishaps, including Cindy's run-in with a feral cat, Ray's botched attempt at intimacy with Brenda, Shorty's encounter with a marihuana-fueled monster, and Alex's steamy tryst with a ghostly lover who, much to her surprise, becomes quite...enthusiastic. As the teens stumble upon Professor Oldman's true intentions, they must band together to escape the clutches of Hell House and its unholy inhabitants.

As the professor's lifeless body slumps against the cold stone floor, a chilling specter emerges from the shadows. The vengeful spirit of Hugo Kane, master of Hell House, ensnares all exits, trapping the terrified teens within the labyrinthine manor. Confronted with their own mortality, they turn to Dwight's arsenal of technological devices, designed to harass and torment the otherworldly foe. As the group navigates the treacherous halls, Cindy is stalked by a skeletal apparition, only to be rescued by Brenda, who dispatches the undead interloper with a brutal display of bone-crushing force.

Meanwhile, Shorty finds himself face-to-face with the very entity that claimed the professor's life. Undaunted, he dons a makeshift mask to conceal her grotesque features and indulges in a macabre tryst. Hanson, however, is not so fortunate; he becomes unwittingly possessed by the malevolent force, leaving his friends powerless against its dark influence. Cindy, Brenda, and Theo attempt to exorcise the demon by conjuring a trio of angels, only to suffer a humiliating defeat at the hands of their tormentor.

As the stakes grow higher, the group devises a desperate plan: using Cindy as bait, they lure Kane's ghost into a trap designed to eradicate its malevolent presence. But two months later, the very same Cindy finds herself ensnared in a new predicament. Despite her initial reluctance, she has grown close to Buddy and the parrot he despises, even as they take their daily stroll. However, when Buddy vanishes without warning, Cindy's world is turned upside down by Hanson's unexpected return – only to be crushed beneath the wheels of Shorty's car, which hurtles out of control with the female ghost perched atop it, still concealing her disfigured visage behind a makeshift mask.
