Orphaned brothers Sam (Rudy Pankow) and Nathan “Nate” Drake (Tom Holland) find themselves in a precarious situation as they attempt to pilfer a valuable map originating from the Magellan expedition at a Boston museum. Sam recounts how Magellan, who famously created the first world map, was in pursuit of gold and, tragically, never returned home. This treasure is believed to be hidden, waiting for someone brave enough to uncover it. After being expelled from their orphanage due to his third infraction, Sam slips away to seek his own fortune, promising Nate that he will return, leaving behind a ring that belongs to their ancestor, Sir Francis Drake.

Fast forward fifteen years, and Nate is now a bartender in New York City, where he also has a knack for pickpocketing wealthy customers. Enter Victor “Sully” Sullivan (Mark Wahlberg), a seasoned fortune hunter who once collaborated with Sam in pursuit of treasure hidden by the crew of Magellan. Sully reveals to Nate that Sam mysteriously disappeared after aiding him in the theft of Juan Sebastian Elcano’s diary. Legend has it that Magellan never completed his journey around the globe; instead, he met his end on a Philippine beach. The diary, crafted by Elcano and his fellow crew members, indicates that their expedition was not financed by the King of Spain but rather by the House of Mancada, containing a fortune estimated at $5 billion.

Unsure of Sam’s fate, especially after receiving no further postcards from him, Nate decides to team up with Sully to search for the treasure, believing that it might lead him to his long-lost brother. Their quest takes them to an auction where they attempt to steal a golden cross associated with the Magellan crew. Here, they cross paths with Santiago Moncada (Antonio Banderas), the last heir of the Moncada family, and Jo Braddock (Tati Gabrielle), the head mercenary for Moncada. As Nate is ambushed by Braddock’s forces, a fierce fight ensues, providing the perfect distraction for Sully to make his move and seize the cross.

The duo travels to Barcelona, where it is said the treasure lies concealed, and reconvenes with Sully’s associate, Chloe Frazer (Sophia Taylor Ali), who possesses another cross. According to Elcano’s diary, Magellan did indeed discover a wealth of gold, which the crew buried in Barcelona, subsequently misleading the Moncada family into believing the expedition was a failure. In a disturbing turn of events, Moncada, upon learning that their family fortune is about to be donated, orders Braddock to eliminate his father, seeking to claim the wealth for himself.

Following the clues from Elcano’s diary, Nate, Chloe, and Sully venture to Santa Maria Del Pi, uncovering a hidden crypt behind the altar. Once inside, they discover a trapdoor that floods the crypt with water as they attempt to open it. Sully comes to their aid, fighting off another ambush led by Braddock. With the help of the two crosses, they unlock a secret passage, unveiling a map that directs them to the Philippines as the location of the treasure. However, Chloe betrays Nate, taking the map to Moncada and subtly indicating that Sully might be concealing crucial information about Sam.

Sully reveals that after he and Sam successfully recovered the diary, they were ambushed by Braddock, resulting in Sam being shot, while Sully barely escaped. Moncada, Chloe, and Braddock’s team board a cargo plane to pursue the treasure, only for Braddock to turn on Moncada, killing him and seizing control of the operation. Nate and Sully cleverly stow away in Moncada’s Mercedes car to confront Braddock mid-air.

A chaotic skirmish breaks out; while hiding from Braddock’s henchmen, Chloe is discovered, leading to an all-out brawl. Sully parachutes from the plane with the invaluable map, leaving Nate and Chloe to plummet into the Philippines. They eventually piece together the map’s true coordinates using clues Sam had left for Nate in earlier postcards, which instructs him on how to use the keys as a compass. This leads them to the exact burial site of the treasure, though Nate cleverly gives Chloe false coordinates, unsure of her loyalty. Discovering the long-lost Magellan ships, Nate is reunited with Sully just as Braddock’s team closes in.

In an exhilarating escape, Sully commandeers a helicopter while Braddock’s men engage Nate and local militias. The tension escalates as Nate skillfully shoots down a pursuing helicopter with one of the ship’s cannons. As the battle reaches a climax, Braddock tries to thwart Nate’s progress by dropping the ship’s anchor, leading to her ultimate demise as the ship falls on her. With Philippine naval units closing in, Nate and Sully manage to snag a few pieces of treasure while Chloe is left empty-handed, deciding to follow the duo instead.

In a post-credits scene, Nate encounters an intermediary working for Roman. He offers the ring from his ancestor, seeking a “Nazi map,” only to be met with an attempt to betray him. Just in time, Sully intervenes, saving Nate as they narrowly escape, though they find themselves captured by an unknown figure waiting in the shadows.
